The speaker stops working when you try to listen to something important it can either be related to some hardware issue or it could be related to software. Check iPhone Volume and Silent Mode

When you have connected your iPhone 6s to the car device via Bluetooth speaker then it might be the reason your iPhone 6s speaker not working. Make sure you turn off the Bluetooth if you paired your phone with other device. You can turn it off by going to Settings>Bluetooth.
Update the iOS
The updating of the software of your iPhone 6s sometimes resolves the speaker problem. Try updating the iOS as there is no harm in updating your iPhone. To update the software you need to go to Setting>General>Software Update. If the software update is available tap Install Now. Before installing the update for your phone make it sure you have about 1.5 GB of free space in your phone.

- Connect your iPhone to your PC using the cable.
- Launch iTunes.
- Go to the summary tab, and click Restore iPhone.
- Let the restoring process to finish then setup your iPhone again.
